Surface Plasmon Resonance

Advantages: • Assess real-time binding (kinetics)

• Provides information on relative binding affinity, concentration and isotype. • Enables detection of low-affinity antibodies (not detectable by other assays) and all isotypes Disadvantages: • Expensive, dedicated equipment; single vendor • Immobilisation of therapeutic could mask epitopes (e.g., amine coupling vs aldehyde coupling) • Regeneration step is difficult to optimize (may degrade immobilised therapeutic) • May be less sensitive than other binding assays
