Electrochemiluminescence (ECL)

MesoScale Discovery (MSD) platform: based on a technology process that use a label designed to emit light when electrochemically stimulated.

The MSD platform has gained significant traction in industry for immunogenicity assessment.

Electrochemiluminescent detection molecule: sulfo-TAG label - chelate of ruthenium (II) and tris-bipyridine that emits light at 620 nm following a voltage-stimulated oxidation-reduction process. Stable, non-radioactive molecule, MW of ~1000 Da.

MSD Platform - Pre-Coated Plates

Mix sample / control Ab , biotinylated therapeutic , & sulfo-TAG therapeutic .

Rely on the multivalent nature of the ADA, to bind both capture and detection reagent (e.g., therapeutic) hence forming a bridge

Transfer to streptavidin coated plates

Add read buffer (TPA) Measure ECL counts
