2. 10-20 or 3-6/sex/group = 80 or 24 animals 3. Treat oral, diet, iv, im, 2-39 weeks 4. 3 dose levels drug and 1 control group 5. Highest dose should cause toxicity, lowest  therapeutic 6. Record response to treatment, clinical condition etc. 7. Toxicokinetics, clinical. chemistry, haematology, urinalysis 8. Post-mortem & histopathology 20-40+ tissues
