Ready-to-use cells vs Continuous cell culture Continuous cell culture: • Requires weekly resource for maintaining cells in culture. • Contamination of cultures mean downtime to get back up to assay Ready to use: • a single uniform large bank of e.g. 200 vials prepared and frozen so as to use e.g. 1 vial per assay. • Storage space for many more vials required initially • allows confidence that assay cells will not change response at older passages. • Not all cells respond to thaw and use conditions.

• Reporter gene assays – offer higher level of sensitivity, specifically engineered for signal transduction of specific receptors, also for multi-cell type assays ADCC etc

• Automation – can be labour intensive initially to understand programming of instruments, in the long term increases productivity and consistency e.g. liquid handlers FeliX, Echo.

• Ready to use cells – Cells banked as a group of vials all at the same “age”/passage and density.
